How it works
Each shared reagent is per-reaction volume × (samples + extra reactions). The extra reactions are not waste: pipetting loses a little at every transfer, and a mix made for exactly the number of tubes runs out on the last one.
Template DNA is the exception. It goes into each tube separately, so it is not part of the mix you share out — it is listed per tube and left out of the master mix total. Getting that wrong is the usual reason a calculated mix does not match what ends up in the rack.
Worth checking
The default reagents are an ordinary 25 µL reaction. Change the per-reaction volumes to match your own protocol rather than trusting somebody else’s — a master mix is only right for the reaction it was written for.
